HEART RATE VARIABILITY DURING nNOS INHIBITION IN SPONTANEOUSLY HYPERTENSIVE RATS

چکیده
PURPOSE: The current study investigated role of nitric oxide (NO), produced by neuronal nitric oxide synthase (nNOS) on heart rate variability (HRV) in spontaneously hypertensive rats (SHR). METODS: The selective neuronal nitric oxide synthase (nNOS) inhibition was performed by application of 7-Nitroindazole (7NI) in conscious normotensive Wistar rats (n=10) and in SHR (n=10), intravenously in a dose 2mg/kg/h. Blood pressure wave was registered directly through a femoral artery catheter in a control period and during nNOS inhibition. In spectrograms of heart rate, derived by FFT algorithm, spectral power (P) in the low (LF), mid (MF) and high (HF) frequency band were studied. Throughout relation, PMF/PHF the sympatho-vagal balance was determined. RESULTS: The nNOS derivate NO in Wistar rats decreased PMF from 1.39±0.34 to 0.43±0.03 ms, p<0.05 and altered PMF/PHF ratio from 0.80±0.1 to 0.44±0.03, (p<0.05). In SHR, nNOS inhibition did not affect HRV. CONCLUSIONS: The absence of inhibiting action of nNOS-derived NO on sympathetic neurotransmission in normotensive rats led to a decrease of sympathetically mediated oscillations in heart rate and to displacement of sympatho-vagal balance. The lack of effect of nNOS-derived NO inhibition in SHR on the HRV may be a result of the impaired nNOS signaling pathway in SHR.

Changes of Plasma Renin Activity after Selective Nnos Synthase Inhibition in Spontaneously Hypertensive Rats
PURPOSE: The present study investigates the role of nNOS in the regulation of plasma renin activity in spontaneously hypertensive rats (SHR). METHODS: Experiments were carried out on conscious normotensive Wistar rats and on SHR. One day before experiment femoral artery for a direct arterial blood pressure (ABP) measurement, femoral vein for drug application and bladder for urine collection wer...
متن کاملConverting enzyme inhibition normalizes QT interval in spontaneously hypertensive rats.
We quantified the repolarization time (so-called QT interval) in a rat, an animal species that does not show a well-characterized T wave on surface ECG. We used spontaneously hypertensive rats (SHR) and converting enzyme inhibition to demonstrate a reversible increase in QT interval in pressure-overloaded hearts in the absence of ischemia. An implanted telemetry system recording ECG data in fre...
متن کاملLisinopril indifferently improves heart rate variability during day and night periods in spontaneously hypertensive rats.
The aim of this work was to investigate the effect of 10 weeks of lisinopril treatment to spontaneously hypertensive rats (SHRs) on day/night variations of blood pressure, heart rate and autonomic cardio-regulation parameters. Male SHR with surgically implanted radio-telemetry implant that provided direct measurements of arterial pressure and electrocardiogram wave were used. Animals were alloc...
متن کاملNoradrenergic cell specific gene transfer with neuronal nitric oxide synthase reduces cardiac sympathetic neurotransmission in hypertensive rats.
Nitric oxide-cGMP pathway can inhibit cardiac norepinephrine (NE) release. Sympathetic hyper-responsiveness in hypertension may result from oxidative stress impairing this pathway. We tested the hypothesis that the gene transfer of neuronal NO synthase (nNOS) could restore sympathetic balance in the spontaneously hypertensive rat (SHR). An adenovirus (5x10(10) particles) constructed with a nora...
متن کاملHemodynamic, Morphometric and Autonomic Patterns in Hypertensive Rats - Renin-Angiotensin System Modulation
BACKGROUND Spontaneously hypertensive rats develop left ventricular hypertrophy, increased blood pressure and blood pressure variability, which are important determinants of heart damage, like the activation of renin-angiotensin system. AIMS To investigate the effects of the time-course of hypertension over 1) hemodynamic and autonomic patterns (blood pressure; blood pressure variability; hea...
